Influence of Interleukin-10 promoter polymorphisms |
Results from the CAPRISA 002 Acute Infection study, investigating the influence of Interleukin-10 promoter polymorphisms on susceptibility and primary HIV-1 pathogenesis was published in the Journal of Infectious Diseases earlier this year. The Interleukin-10 (IL-10) proximal promoter region consists of 2 known bi-allelic polymorphisms, which are related to levels of IL-10 production. Polymorphisms resulting in decreased production have been shown to increase likelihood of HIV-1 acquisition and accelerate CD4 decline. This has led to the suggestion that IL-10 may protect against disease progression. However, more recent animal models with lymphocytic choriomeningitis virus have suggested that removal of IL-10 enhanced T cell immune responses, development of antiviral memory and viral clearance.

Results from the CAPRISA 002 Acute Infection Study |
Results from the CAPRISA 002 Acute Infection Study, investigating the role of neutralizing antibodies in neutralization escape of subtype C virus, was published recently in PLoS Pathogens. In most HIV-1 infected individuals neutralizing antibodies that target the Env region of the virus develop and reach detectable levels in the first few months of infection and subsequently block viral entry. It is presumed that the pressure exerted by autologous neutralizing antibodies may account for much of the variation that occurs in Env during early infection. In both HIV-1 subtype B and SIV, neutralization escape has been documented with contemporaneous viruses showing less sensitivity to autologous neutralization than the earlier virus. In subtype C infection however, the autologous neutralizing antibody response appears to differ somewhat to the subtype B virus and is less well characterized.
